Luthor had theorized that because Superman's powers first manifested themselves when he was a teenager and evolved more as he grew older that they were tied to his sexual development. If he could just get a sperm sample and synthesize its potency into a serum he would be able to change himself into "super" being. Obviously there was no easy way to obtain any semen from Superman--it would have to be without his knowledge. Luthor needed to approach Superman in his secret identity. Obtaining proof that Superman was Clark Kent turned out to be remarkably easy & once he knew where he lived & slept he could begin phase one.
The kryptonite meteorite that Luthor had stolen months years provided Luthor with the meas to carry out his plan. Having broken in to Kent's apartment he sprinkled a powder of pulverized kryptonite on Kent's bedroom pillow knowing that in this small quantity it would put him into a deep sleep.
Returning to the bedroom that night Luthor indeed found Superman out cold.
